Immunogen: The original hybridoma was generated by fusing anti-Dengue antibody producing, EBV immortalised B-cells from a DENV-immune patient, with HMMA2.5 myeloma cells.
Specificity: This antibody binds to Dengue virus 3 envelope protein. A single mutation in the DI-DII hinge (lysine insertion between Q269 and N270) abolishes binding. Fibriansah et al (2015, PMID: 25698059) report that a 5J7 Fab binds across three envelope proteins and engages three functionally important domains, each on a different protein.
Application Notes: This antibody binds to DENV3 E-protein by ELISA and flow-cytometry. It has also been shown to neutralize DENV3 virus, but not DENV1, DENV2 or DENV4.
