Immunogen: mAb ZKA185 was selected from EBV-immortalized memory B cells derived from ZIKV-infected, DENV-naïve human donors based on its ability to neutralize ZIKV infection.
Specificity: This antibody is able to specifically neutralize Zika virus, however it does not react with recombinant envelope protein (E-protein) or EDIII. As a result ZKA185 has been named a 'Neutralizing Non-Binder' (NNB) . NNBs interact with quarterny epitopes displayed on infectious virions but not soluble proteins. It is suspected that the epitope ZKA185 lies within the the EDE region (i.e. the E-dimer dependent region), however research into this is still ongoing. No neutralization of or binding to Dengue virus could be detected.
Application Notes: This antibody, in the form of a human IgG1, has been shown to specifically neutralize Zika virus in vitro (strains tested: H/PF/2013 &MR766).
